From the gui, if I push live a few times and switch channels, then the light comes through. When I see light on manual, I can run my script and it images fine.

If you have the possibility to use the ethernet interface you can get a graphical interface that for example shows if the light engine is in standby. This makes it easy to ensure that the light engine is active before you start.

You can also reset the standby timer without illuminating your sample from micro-manager by setting all intensities to zero, turning on one channel and then either snapping an image or flipping “State” from zero to one and then back again. Wait a couple of seconds for the light engine to warm up and then you are ready to image. We also had problems with too infrequent imaging and then used this approach every four minutes to make sure that the light engine never goes into standby.

At least on my light engine, the warm up problem does go away if you wait. Since you state you are writing your own imaging scripts you can catch the exception when it occurs, wait for a while, and then snap again. Here is a beanshell code example:

The beanshell script supplied by Lumencor does work, but maybe not the way you expect them to. If you want to use them you have to connect to the light engine using the FreeSerialPort device adapter. The downside with this is that you can’t control the light engine, using the Lumencor device adapter connected to the COM-port (you can’t have two device adapters connected to the same com-port). On the other hand, if you use the ethernet interface you can actually get both at the same time. Use the Lumencor device adapter together with the ethernet interface (just type in the ip-number of the light-engine instead of the COM-port when you add the device to micro-manager). At the same time, also add an FreeSerialPort device to micro-manager using COM4. Now you can use the bean shell scripts that Lumencor supplied you on COM4, while still being able to control the light-engine using the standard Lumecor device adapter over ethernet. Note that I have set it up like this, and it it works for a couple of snaps, but I haven’t done any extensive testing.

The company told me the laser sleeps if idle for 5 minutes. There’s a serial command, “WAKEUP” but I’m having trouble sending it to the serial port. They sent me this example beanshell script to wake the light engine:

Problem is that Micro-Manager does not know about COM4. The Lumencor driver talks to the COM port directly, i.e. the COM port is not a device within Micro-Manager that you can communicate with. Lumencor will need to add a wake up call to their driver / device adapter.

If you wait a little, does the error go away? It sounds like the Lumencor light engine wants to warm up the lasers before doing anything. If the issue is not manageable, I’d suggest contacting Lumencor, or the system integrator that sold you the system.
