Acquire White Reference Image: Use a slide that does not have sample or dirt on it. Adjust the exposure so that no pixels saturate. Create the Shading Reference Image: Use the Arithmetic dialog (subtraction mode) to do Shading = White Reference – Background Reference. Acquire Specimen image. Be sure the camera does not clip or saturate. If the camera does, add neutral density filters or decrease exposure time, and re-acquire the white reference image. If you are acquiring multiple images you may want to acquire them to a single stack, rather than separate image windows. Save the image(s) to disk before experimenting with shading correction for the first time. Not mentioned in the tech note is that MetaMorph users can activate the "ODscaled" dropin from MMAdmin. This enables calculating optical density for each pixel in an image. It is crucial that the microscope be optimized to not have glare -- set the condenser numerical aperture diaphragm correctly (and consistently for the entire data set. After all these years I had no idea SNR for filters was calculated that way. Seems kind of silly. I don't understand why signal intensity is somehow related to filter bandwidth. Max intensity is max intensity regardless of how broad or narrow the filter is!
The second filter transmits all of the incident radiation within a 10 nm narrow passband and blocks to a level of OD6 within the remaining 990 nm. Please note that the OD level is the same as for the first filter! Only the passband is ten times more narrow. In this case the SNR calculation yields 40 dB, which seems to indicate that the filter is not as good as the first one. But this is not the case! The blocking performance is the same but the narrower band lets pass 10 times less light. This effect should be considered when specifying optical filters.

SPW501 Spanner WrenchThe double-ended SPW501 Spanner Wrench is designed for tightening Ø1" RS posts with torque slots using the RS Post end of the wrench, as illustrated in the photo to the right. It can also be used to tighten Ø1/2" TR posts and Ø1" RS posts with through holes using the TR Post end of the wrench. The SPW403, SPW401, and SPW402 spanner wrenches are designed for use with our locking collars for M2.5 x 0.20, 3/16"-100, and 1/4"-80 adjuster screws, respectively. The SPW403 can also tighten or loosen the Zθ locking collars on our FiberPort collimators. The spanner wrenches are 0.75" (SPW403) or 1.50" (SPW401 and SPW402) long, have a knurled grip, and are constructed of 303 stainless steel. The hollow construction allows an adjuster to pass through the entire length of the spanner wrench.
Thorlabs' Adjustable Spanner Wrench features a spanning range that is continuously adjustable from 0.12" to 2.89" (3.0 mm to 73.4 mm). It is capable of providing an exact fit for retaining rings as large as the SM3RR for Ø3" Lens Tubes and Mounts. The diameter spanned by the blades remains centered while the knurled knob is adjusted.
An optical density of six (or OD6) for example describes that at a given wavelength a filter transmits only one millionth of the incident radiation. Please note that optical density is always calculated from the transmittance value on a scale from 0 to 1, not from the %-value that is listed for correspondence in the table above.

Let us look at two bandpass filters in order to exemplify how optical density and SNR are related. The first filter transmits all of the incident radiation (T=1) within a 100 nm wide passband and blocks to a level of OD6 within the remaining 900 nm. As the name suggests SNR is the ratio of the (desired) signal to the (undesired) noise. For comparison with the filter's OD, we calculate SNR for a detector (with constant spectral sensitivity) that is illuminated through the filter with a broad-band light source (of constant spectral intensity) by calculating the integral below the red curve. SNR is often given in decibel, in this example the sensor has an SNR of 50 dB.

For retaining rings not covered by these cylindrical spanner wrenches, please see the SPW801 Adjustable Spanner Wrench sold at the bottom of the page. The SPW801 features an adjustable spanning range as well as long blades that extend 1.6" (40.7 mm) from the body of the wrench. Hence, for lenses with a high radius of curvature, we recommend using the SPW801 over the cylindrical spanner wrenches, whose guide flanges may come into contact with the lens.

Spanner Bits allow SM-threaded retaining rings to be installed or adjusted without damaging optics that may be in place. They feature a standard 1/4" hex shank that allows them to be used with a manual driver handle, such as the one included in the kit below.
The reversible blades have a flat tip on one end intended for use with retaining rings, and an edge-offset fine point on the opposing end for adapters with holes. The blades extend 1.6" (40.7 mm) from the body of the wrench. Hence, compared to our cylindrical spanner wrenches sold above, the SPW801 is ideal for securing lenses with a high radius of curvature because the guide flanges on the cylindrical spanner wrenches may come into contact with the lens. In our daily work as filter manufacturer we often talk to customers with an electrical engineering or imaging background. In these and other fields SNR is an important quantity to describe system or detector performance. Filters on the other hand are described by transmission and rejection performance at specific wavelengths over a wider wavelength range. Transmission is often given on a scale from 0 to 1 (preferably) or on a %-scale. Rejection is often described as optical density.
We also offer stainless steel spanner bits with a 1/4" hex shank for our SM05, SM1, SM1.5, SM2, and SM3 standards similar to our standard spanner wrenches. A kit is also offered that includes spanner bits for the SM05, SM1, and SM2 standards; a driver handle with a 1/4" hex socket; and a case. The SM1.5 and SM3 spanner bits are not included with this kit.
If using these bits with a power drill, care should be taken to avoid overtightening the retaining ring due to the added torque created by the tool. Electric tools should be used at a low speed, and we recommend that the final tightening of the retaining ring be done by hand.

The field of view is often quite limited, and the front lens element of the objective is placed close to the specimen with which it must lie in optical contact.
Each bit includes guide flanges that prevent most optical elements from being scratched while the bit is being inserted. When using lenses with high curvature, we recommend the SPW801 Adjustable Spanner Wrench sold below, which uses long blades that prevent scratches caused by the guide flanges.
