Principle of microscope

This grant covers costs related to your attendance to the course (registration, travel and accommodation costs). The grant is restricted to PhD students and postdocs who conduct basic biomedical research.Whether you are eligible to apply for a travel grant, depends on when you received your university entrance qualification (e.g. Abitur, A-Levels, High School Diploma, Final State Examination):

Diaphragm or Iris: Many microscopes have a rotating disk under the stage. This diaphragm has different sized holes and is used to vary the intensity and size of the cone of light that is projected upward into the slide. There is no set rule regarding which setting to use for a particular power. Rather, the setting is a function of the transparency of the specimen, the degree of contrast you desire and the particular objective lens in use.

The topics will include fluorescence microscopy techniques such as fast time-lapse microscopy of living cells, spinning disk and point-scanning confocal microscopy of large (3D) samples, photo-manipulation as a tool, multicolour single-molecule localization microscopy (SMLM) with spectral demixing, multiplexed imaging, expansion microscopy, FRET-based techniques. For demanding applications, we will cover the aspects of image processing and analysis and the possibilities of microscope automation. The modules will cover all necessary knowledge and demonstrate tools to conduct similar pipelines at your home lab enabling you to answer emerging biological questions.

There is the possibility to apply for a childcare grant to offset child care costs incurred by participants, speakers, trainers and organisers when attending a course. Eligible costs include (but are not limited to) fees for a babysitter or child-care facility and travel costs for a caregiver. Please note that priority will be given to early-stage researchers. There is a limited amount of funding available for the childcare grants and funds will be distributed amongst eligible applicants.

The course is limited to 16 participants. For selection purposes, please note that your application will not be considered without a letter of motivation.

If you are travelling to the conference within Germany then you are eligible for the Deutsche Bahn ‘Event Ticket’ (called the ‘Veranstaltungsticket’ in German). This will result in a lower ticket price if your travel distance to Heidelberg is more than 100 km. You need to provide proof of your event attendance when purchasing the ticket.

Condenser Lens: The purpose of the condenser lens is to focus the light onto the specimen. Condenser lenses are most useful at the highest powers (400x and above). Microscopes with in-stage condenser lenses render a sharper image than those with no lens (at 400x). If your microscope has a maximum power of 400x, you will get the maximum benefit by using a condenser lenses rated at 0.65 NA or greater. 0.65 NA condenser lenses may be mounted in the stage and work quite well. A big advantage to a stage mounted lens is that there is one less focusing item to deal with. If you go to 1000x then you should have a condenser lens with an N.A. of 1.25 or greater. All of our 1000x microscopes use 1.25 Abbe condenser lens systems. The Abbe condenser lens can be moved up and down. It is set very close to the slide at 1000x and moved further away at the lower powers.

Magnification: Compound microscopes are designed for higher magnifications, typically used for observing microscopic details. Other microscopes may have lower magnification capabilities, suitable for larger specimens or samples.

Illuminator: A steady light source (110 volts) used in place of a mirror. If your microscope has a mirror, it is used to reflect light from an external light source up through the bottom of the stage.

Types of microscope

Sample Size and Depth of Field: Compound microscopes are designed to observe thin, transparent specimens placed on glass slides. They offer a narrow depth of field, allowing clear focus on one plane at a time. Other microscopes, like stereo or electron microscopes, can accommodate larger specimens or samples with more depth, providing a wider depth of field.

Eyepiece/Ocular: Compound microscopes commonly have a pair of eyepieces that provide binocular vision. Other microscopes may have a single eyepiece or sometimes no eyepieces at all.

In most places the electricity is 220 volts AC (50 cycles). An adaptor and a plug that fits the German socket may be needed for your appliances/laptop (i.e. American, Japanese, etc.). A USB charging station for electronic devices is available at the registration desk..

All meals and coffee breaks are included in the registration fee. Our catering staff will prepare a wide variety of vegetarian meals, meat and fish dishes, soups, pasta, fresh fruit and vegetables, as well as a variety of desserts.

The registration fee should be paid only after acceptance to the course. The results will be announced approximately 2-3 weeks after the application deadline.

Accommodation is not included in the registration fee. Participants are expected to book and pay for their own accommodation.

During the event, we provide conference shuttle buses to and from EMBL. In addition, there is the public bus 39A that serves the EMBL campus and taxis can be easily booked at any time. Applications: Compound microscopes are commonly used in fields such as biology, medicine, and research, where detailed examination of small structures is required. Other microscopes, such as stereo microscopes, are utilized for examining larger objects or conducting dissections. Electron microscopes are used for high-resolution imaging of nanoscale structures.

The scientific organisers will select the recipients of registration fee waivers during the participant selection process for courses. Results will be announced approximately 6 – 8 weeks before the event start date, however for some events this may be delayed. How to Focus Your Microscope: The proper way to focus a microscope is to start with the lowest power objective lens first and while looking from the side, crank the lens down as close to the specimen as possible without touching it. Now, look through the eyepiece lens and focus upward only until the image is sharp. If you can't get it in focus, repeat the process again. Once the image is sharp with the low power lens, you should be able to simply click in the next power lens and do minor adjustments with the focus knob. If your microscope has a fine focus adjustment, turning it a bit should be all that's necessary. Continue with subsequent objective lenses and fine focus each time.

Objective Lenses: Usually you will find 3 or 4 objective lenses on a microscope. They almost always consist of 4x, 10x, 40x and 100x powers. When coupled with a 10x (most common) eyepiece lens, total magnification is 40x (4x times 10x), 100x , 400x and 1000x. To have good resolution at 1000x, you will need a relatively sophisticated microscope with an Abbe condenser. An Abbe condenser is composed of two lenses that control the light that passes through the specimen before entering the objective lens on the microscope. The shortest lens is the lowest power, the longest one is the lens with the greatest power. Lenses are color coded and if built to DIN standards are interchangeable between microscopes. "DIN" is an abbreviation of "Deutsche Industrial Normen". This is a German standard that has been adopted internationally as an optical standard used in most quality microscopes. A typical DIN standard microscope objective lens has a 0.7965" (20.1mm) diameter threads, 36 TPI (threads per inch), and a 55º Whitworth. Many high power objective lenses are retractable (i.e. 40XR). This means that if they hit a slide, the end of the lens will push in (spring loaded) thereby protecting the lens and the slide. All good quality microscopes have achromatic, parcentered, parfocal lenses.

Single rooms have been reserved for 6 nights from Saturday, 17 May 2025 to Friday, 23 May 2025 at the ISG hotel (Im Eichwald 19, 69126 Heidelberg).

This practical course will cover advanced light microscopy techniques and participants will learn how to derive qualitative and quantitative insights on molecular mechanisms in mechanisms in cells, tissues and developing organisms. Invited guest researchers together with microscopy experts from EMBL will foster an intense information flow with a balance of lectures and practical workshops. The focus of the course will be on the use of fluorescence microscopy to obtain information about localisation and functions of proteins and other bio-molecules at different spatial scales.

Stage with Stage Clips: The flat platform where you place your slides. Stage clips hold the slides in place. If your microscope has a mechanical stage, you will be able to move the slide around by turning two knobs. One moves it left and right, the other moves it up and down.

Microscope parts

This course is directed towards researchers in the life sciences with access to light microscopy equipment, who have an immediate need to apply the techniques they will learn to solve biological problems at the cellular and molecular level in vivo. As the course will be primarily focused on the advanced techniques, we expect participants to have basic knowledge of light microscopy.

Childcare grants will be allocated in the same timeframe (6-8 weeks before the event start date). Please note that priority will be given to early-stage researchers.

The travel grant will cover the cost of travel (airfare, train, bus, taxi, accommodation, visa, and/or registration fees*) and is provided up to specified caps which are normally as follows:

In your application you will be asked to summarise your current work,  answer questions regarding why your lab cannot fund your attendance, and how your attendance will make a difference to your career. Application for financial support will not affect the outcome of your registration application.

Illumination: Compound microscopes often have built-in illumination systems, such as a substage light source, condenser, and diaphragm, to provide transmitted light through the specimen. Other microscopes, like dissecting or fluorescence microscopes, may utilize different lighting techniques or illumination configurations.

Wi-Fi is available on campus. Log in using the EMBL-Events network and the event specific password, which will be provided on site. The eduroam network (secure, worldwide roaming access service developed for the international research and education community) is also available.

Shuttle buses will go from the ISG Hotel to EMBL and back, mornings and evenings. A bus schedule and location of the bus stops will be made available prior to the meeting.

What is microscope in science

Revolving Nosepiece or Turret: This is the part of the microscope that holds two or more objective lenses and can be rotated to easily change power.

The organisers may reduce the grant cap to accommodate more participants. Recipients will be notified of their travel cap amount when they are informed of the outcome of their application. Original receipts must be provided with your signature for all costs incurred within two months of completion of travel. Scanned copies cannot be accepted.

During the conference, an EMBL Photographer may be taking photos. If you would not like to appear in these, please inform the photographer or a member of the Course and Conference Office.

Historians credit the invention of the compound microscope to the Dutch spectacle maker, Zacharias Janssen, around the year 1590 (more history here). The compound microscope uses lenses and light to enlarge the image and is also called an optical or light microscope (versus an electron microscope). The simplest optical microscope is the magnifying glass and is good to about ten times (10x) magnification.

Objective Lenses: Compound microscopes have multiple objective lenses mounted on a rotating nosepiece, typically with magnifications ranging from 4x to 100x or higher. Other microscopes, such as dissecting or stereo microscopes, usually have fixed magnification lenses.

Rack Stop: This is an adjustment that determines how close the objective lens can get to the slide. It is set at the factory and keeps students from cranking the high power objective lens down into the slide and breaking things. You would only need to adjust this if you were using very thin slides and you weren't able to focus on the specimen at high power. (Tip: If you are using thin slides and can't focus, rather than adjust the rack stop, place a clear glass slide under the original slide to raise it a bit higher).
