Light sheet fluorescence microscopy recently became a popular technique in developmental biology. This method allows for long-term imaging of many different samples with fast acquisition and most importantly, very low phototoxicity. However, so far most systems were custom-made and not available to the majority of biologists. This recently changed, when commercial systems entered the market. Here, we present a protocol explaining how to best make use of such systems and how to deal with the big amount of data that is generated with light sheet experiments.

Overall, this protocol shows the cross disciplinary nature that is important when acquiring light sheet  data  as  developmental  biology  needs  computer scientists. With this hands-on manual it should also be easy for first time users to get over initial reserve and start imaging in order to ask the questions in developmental biology that were much more difficult to ask previously.

Another thing we like about this drive-thru is that the lights are set on a dark backdrop. We felt that this made the lights extra bright and fun. Usually lights are standing alone, but on the dark background, the lights standout no matter the weather and time of day.

Our open access video protocol (Icha, Schmied et al., 2016) establishes a versatile light sheet microscopy experimental pipeline. This pipeline is based on a commercial microscope and an open source software solution for data processing. Specifically, we used the Lightsheet Z.1 microscope from Zeiss and the Multiview reconstruction application in Fiji to process the data. The implications of this approach are demonstrated by imaging several stages of retinal development in zebrafish spanning from optic cup formation to neuronal translocation. However, this pipeline can be used for long-term time-lapse imaging of a plethora of morphogenetic events in different model organisms. Overall, the protocol covers the essential steps of a light sheet microscopy experiment from mounting the sample to processing the data. It needs to be noted though that the most complicated step in light sheet microscopy is not acquiring the images but the subsequent read out of the information in the data, especially, when multiple views are imaged and need to be fused. The solution provided in the protocol is to embed fluorescent beads as fiduciary markers around the sample to register the different views onto each other, which allows the reconstruction of the entire imaged volume of the sample. For fast and interactive visualization of the often very large datasets we are using the BigDataViewer in Fiji.

World of Illumination is a light show in the Salt Lake Valley. This event is held at the Utah State Fairpark and runs during the holidays. The cost to drive through the light show is a little higher than some of the other popular light drives, but we found the cost to be worth it. World of Illumination was definitely a new favorite.

HFSP promotes and participates in several events every year. We are committed to bringing together the scientific community and forging new opportunities to network and have scientific discussions that help to create bridges and partnerships for the future of frontier science.

There were several things that we liked about World of Illumination. First, the lights were thick, and the bulbs were big. This really makes the lights pop. As you drive back and forth, the lanes are narrow so you really get the feeling that you are right in the show. Finally, the lights are higher up. There are multiple tunnels and displays that cross the road, so you feel surrounded by lights throughout the entire drive. We’ve never really seen a show that makes you feel that you are right in the lights like World of Illumination does.

World of Illumination puts on Christmas light shows throughout the United States. Each show is based on a different theme. Salt Lake City’s theme is Reindeer Road in 2024 (It was Arctic Adventure in 2022 and Candy Rush in 2023), so all of the lights are based on the theme. There are arctic animals like polar bears, penguins, seals, and more. And this year (2023) everything is about candy. You can see a mix of pictures from the last few years.

To truly understand how development proceeds and how cells form tissues, it is very important to observe cell and tissue biology when it happens and where it happens in the unperturbed embryo. However, it becomes more and more clear that confocal systems, that have been widely used for such studies come with certain limitations, one of the major ones being the induction of phototoxicity. To this end, the development of light sheet microscopy was an important step to reduce this problem and allow for long, fast and gentle imaging of many tissues in many different organisms. One of the organisms best suited to light sheet imaging is the translucent zebrafish embryo. Organ development is often conserved between the zebrafish and higher vertebrates, making it an ideal model to understand cell biology and tissue mechanics of these processes that can later be transferred to other less accessible systems or ex vivo models. However, one obstacle that needs to be overcome to generate meaningful conclusions from the data is to manage the immense data amounts that result from light sheet experiments. Our protocol deals on one hand with the details of setting up an imaging experiment and on the other, makes suggestions how to analyze the resulting data.

The drive through World of Illumination is quite long, and the music is very well timed to the dancing lights. We also liked the uniqueness of the displays. Spinning snowman, dancing candy canes, and many other fun displays that fit the theme make this a fun drive-through light show. We are excited this has become an annual Christmas light show here in Utah.

It is cheaper to visit on weekdays and before the middle of December, so if you can make that work you can save some money. In 2023, they had tickets at Costco for $35 and they were any day tickets, so keep an eye out this year for that deal. This is the best deal that I have seen.
